In February, 1941, McGuire unsuccessfully tried to join the Eagle Squadron, an American corps of volunteers in the R.A.F. He then integrated the US Marine Corps on July 12th. He began his training at Corsicana Field (Texas) and flew for the first time solo on August 5th. At the end of September, he flew North American BT-9 and AT-6 at Randolph and Kelly airfields, near San Antonio, where he met his future wife Marilyn Giesler.

On June 6th, Mc Guire became First Lieutenant and then on July 20th, he was transferred to the 475th FG created by general Kenney of the 5th AF and assigned to the 431st FS leaded by the Captain Franklin Nichols. His aircraft was the P-38H-1 s/n 42-66592 which he named "Pudgy", his wife's nickname. On August 18th, 1943, during an escort mission of B-25s over Wewak (New-Guinea), he faced for the first time the Japanese fighters : 5 Kawasaki Ki-61s of 68th Sentai and 16 Nakajima Ki-43 of 24th and 59th Sentai (some of them were confused with Zeros). When back from the mission, Mc Guire claimed 2 Zeros, 1 Ki-43 and 1 Ki-61. A Ki-43 was attributed to the Lieutenant Lutton, who had also fired the plane. On August 21st, Mc Guire damaged a Kawasaki Ki-45 of the 13th Sentai and shot down 2 Zeros. He then became an ace.

On August 29th, when protecting again B-24 above Wewak, he shot down 2 Japanese fighters but he got touched and landed in emergency at Tsili-Tsili. "Pudgy" was destroyed and replaced a few days later by "Pudgy II", a P-38H-5 s/n 42-66817. On September 28th, he shot down 2 Nakajima Ki-43 always above Wewak. Some time later, he "competed" an Australian Spitfire pilot to prove the qualities of the P-38 and shook so much his aircraft that it became almost unusable.

On October 15th, he took the P-38H-1 s/n 42-66577 of the First Lieutenant Wood Clodfelter to counter-attack Aichi D3A2 Vals of the 582nd Kokutai and Zeros of the 204th Kokutai over Oro Bay. He shot down a Val but could not finish two Zeros due to a lack of ammunitions. On the 17th, the Japanese came back and McGuire took this time the aircraft of his absent leader, the Major Franklin A. Nichols. He shot down 3 Zeros but got touched and had to jump over the sea. Injured, he was rescued by the speedboat PT-152 and was evacuated at Oro Bay hospital and then in Australia. He had at this time 13 victories (8 less than Richard "Dick Bong").

On December 10th, 1943, McGuire became Captain and returned to operations on the 12th. On the 26th, he shot down four dive bombers over Cape Gloucester, but had only three of them approved. The 4th was awarded to a P-47D's pilot of the 36th FS - 8th FG with whom he played cards for the attribution of this victory ! McGuire then jumped to 16 victories (five behind Bong). But Bong then rised to 28 victories in April 12, 1944 before departing for a "propaganda" tour in the U.S.A.

McGuire at that time no longer increased his score but got "PUDGY III ", a brand new and faster P-38J-15 on March 14. The 475th FG then moved to Hollandia. McGuire became the commander of the 431st FS.

On October 14th, Mc Guire joined the 9th FS for a raid over Balikpapan refineries at Borneo : he shot down at this occasion 4 aircrafts but only 3 were granted to him. On November 1st, the 431th FS moved to Tacoblan, at Leyte. When he was about to land, after a 1 300 km flight, Mc Guire was informed about the presence of a Japanese aircraft : he joined the plane, a Nakajima Ki 44 "Tojo" and shot it down. Despite this victory, he stayed nevertheless at 8 victories behind Bong.

3 more victories on November 10th and 12th, 1944 for Mc Guire : a Nakajima Ki-43 "Oscar" and two Mitsubishi J2M "Jake". Bong having shot down 3 aircrafts too, Mc Guire always stayed 8 victories behind him. The competition between both aces continued until Bong's 40th victory, on December 17th : he was then removed from the operations and sent in the USA to receive the Medal of Honor of the Congress. After a crisis of malaria, Mc Guire took advantage of it : on December 25th, he shot down 4 "Zeke" over Clark's field (Manilla) and 3 others the next day at the same place.

At about 7:00 am, they flew over Fabrica airfield ( Los Negros Island) when a "Oscar" of the 54th Sentai flown by Major Akira Sugimoto appeared. Mc Guire fell on him whereas the Japanese rose towards the group to disrupt it. He stressed too much his curve and started a to roll over on the left before diving to the ground. Too low, Mc Guire collided and exploded.

Sugimoto disappeared and another Japanese arrived, a Nakajima Ki-84 "Frank" of the 71th Sentai, flown by Staff-sergeant Mizunori Fukuda. He managed and shot down Rittmayer who crashed near Pinanamaan. The 2 last P-38s returned to Dulag where the announcement of Mc Guire's death sounded like a bombshell. The investigation retained that he rolled over too close to the ground but the hypothesis whether he was shot down by Sugimoto is not totally excluded. This last one, badly damaged, made a belly landing. His body was found riddled with 6 bullets in the breast, probably killed by partisans Filipinos. Fukuda also made a belly landing at Manapla
